The gut microbiota metabolite indole alleviates liverinflammation in mice

ABSTRACT

The gut microbiota regulates key hepatic functions, notably through the production of bacterial metabolites that are transported via the portal circulation. We evaluated the effects of metabolites produced by the gut microbiota from aromatic amino acids (phenylacetate, benzoate, p-cresol, and indole) on liver inflammation induced by bacterial endotoxin. Precision-cut liver slices prepared from control mice, Kupffer cell (KC)-depleted mice, and obese mice (ob/ob) were treated with or without LPS and bacterial metabolites. We observed beneficial effects of indole that dose-dependently reduced the LPS-induced up-regulation of proinflammatory mediators at both mRNA and protein levels in precision-cut liver slices prepared from control or ob/ob mice. KC depletion partly prevented the antiinflammatory effects of indole, notably through a reduction of nucleotide-binding domain and leucine-rich repeat containing (NLR) family pyrin domain-containing 3 (NLRP3) pathway activation. In vivo, the oral administration of indole before an LPS injection reduced the expression of key proteins of the NF-kB pathway and downstream proinflammatory gene up-regulation. Indole also prevented LPS-induced alterations of cholesterol metabolism through a transcriptional regulation associated with increased 4b-hydroxycholesterol hepatic levels. In summary, indole appears as a bacterial metabolite produced from tryptophan that is able to counteract the detrimental effects of LPS in the liver. Indole could be a new target to develop innovative strategies to decrease hepatic inflammation.

장내 미생물은 중요한 간 기능을 조절하는데, 특히 포털 순환을 통해운반되는 박테리아 대사 산물의 생산을 통해 이루어진다. 우리는 박테리아 내독소 (endotoxin)에 의해 유발 된 간염에 아로마틱 (aromatic) 아미노산 (페닐아세테이트, 벤조에이트, p-크레졸및 인돌)으로부터 장내 미생물에 의해 생성된 대사 산물의 효과를 평가했다. 대조군 쥐, Kupffer 세포(KC)가 결핍된 쥐, 비만 쥐로부터 준비된 정밀 절단 간 조각을 LPS 및 박테리아 대사 산물이 있을 때와 없을 때로 나누어 실험하였다. 우리는대조군 또는 비만 쥐로부터 제조된 정밀 절단 간 조각에서 mRNA 및 단백질 수준 두가지 모두의 염증성매개체의 LPS에 기인한 상향 조절을 용량 의존적으로 감소시키는 인돌의 유익한 효과를 관찰하였다. KC 고갈은 부분적으로 뉴클레오타이드 결합 도메인 및 고류신반복 (leucine-richrepeat; NLR) 계열 피린 도메인 함유 3 (NLRP3) 경로 활성화의 감소를 통해인돌의 항 염증 효과를 일부 예방했다. 생체 내에서 LPS를주사하기 전에 인돌을 경구 투여하면 NF-kB 경로 및 후속적인 전 염증 유전자의 상향 조절의 주요단백질 발현이 감소되었다. 인돌은 또한 간 4b-하이드록시콜레스테롤의증가된 수준과 관련된 전사 조절을 통해 LPS에 기인한 콜레스테롤 대사의 변화를 예방했다. 요약하면, 인돌은 간에서 LPS의해로운 영향을 상쇄할 수있는 트립토판에서 생산된 박테리아 대사 산물로서의 역할을 한다. 인돌은 간염을감소시키는 혁신적인 전략을 개발하는 새로운 목표가 될 수 있다.
