Anti-inflammatory effects of Lactobacillus brevis (CD2) on periodontal disease

Abstract OBJECTIVES: To analyze the anti-inflammatory effects of Lactobacillus brevis extracts on periodontitis patients and to investigate the involved mechanisms in vitro on activated macrophages. METHODS: Eight healthy subjects and 21 patients with chronic periodontitis were enrolled to analyze the effect of L. brevis-containing lozenges on periodontitis-associated symptoms and signs. Before and after the treatment, the patients received a complete periodontal examination. Saliva samples, collected before and after treatment, were analyzed for metalloproteinase and nitric oxide synthase (NOS) activity, immunoglobulin-A (IgA), prostaglandin E 2 (PGE 2) and c-interferon (IFN-c) levels. Arginine deiminase (AD) and NOS activities were determined through a radiometric assay. Metalloproteinases were assayed by zymogram and Western blotting, whereas IgA, PGE 2 and IFN-c were assayed by enzyme-linked imunosorbent assay tests. RESULTS: The treatment led to the total disappearance or amelioration of all analyzed clinical parameters in all patients. This was paralleled to a significant decrease of nitrite/nitrate, PGE 2, matrix metalloproteinase, and IFNc levels in saliva samples. CONCLUSION: Our results suggest that the anti-inflammatory effects of L. brevis could be attributed to the presence of AD which prevented nitric oxide generation. Our findings give further insights into the knowledge of the molecular basis of periodontitis and have a potential clinical significance, giving the experimental ground for a new innovative, simple and efficacious therapeutical approach of periodontal disease. 연구 목적: 치주염 환자에서 Lactobacillus brevis 추출물의 항 염증 효과를 분석하고, 활성화된 대식세포에 대한 체외
진행 관련 메커니즘을 조사하고자 한다.
방법: 8명의 건강한 피험자와 만성 치주염 환자 21명을 대상으로 L. brevis가 함유된 lozenges(마름모꼴 캔디)가
치주염 관련 증상과 징후에 미치는 영향을 분석하였다. 치료 전후에환자들에게 치주 검사를 모두 실시하였다.
치료 전후에 수집된 타액 샘플은 금속단백분해효소(metalloproteinase) 및 산화질소 생성효소
(NOS, nitric oxide synthase)의 활성, 면역 글로불린-A (IgA), 프로스타글란딘 E 2 (PGE 2) 및 c-인터페론 (IFN-c)
수준에 대해 분석되었다. 아르기닌디이미나아제 (AD,Arginine deiminase) 및 NOS 활동은 방사 측정 분석을 통해 결정되었다. 금속단백분해효소는 지모그람(zymogram)과 단백질흡입법(Western blotting)으로 분석된 반면,
IgA, PGE 2 및 IFN-c는 효소결합면역흡착측정법(enzyme-linked imunosorbent assay)으로 분석되었다.
결과: 치료를 통해 전체 환자에게서 분석된 모든 임상 지표에 있어 완전 소실 또는 개선이 이루어졌다.
이것은 타액 샘플에서 아질산염/질산염, PGE 2, 매트릭스 금속단백분해효소 및 IFNc 수준의 유의한 감소 결과가 나온
것과 일맥상통한다.
결론: 우리의 결과는 L. brevis의 항 염증 효과가 산화 질소 생성을 막는 AD(아르기닌디이미나아제)의 존재에 기여할
수 있음을 제시한다. 우리의 연구 결과는 치주염의 분자 기반 지식에 대한 더 깊은 통찰력을 제공하고 잠재적인
임상적 중요성을 가지며, 치주 질환의 새롭고 혁신적이며 간단하면서도 효과적인 치료적 접근법에 대한 실험적 근거를 제공한다.
